How to Read a Peptide Certificate of Analysis: Identity, Purity, and Endotoxin

At a glance: A certificate of analysis, or COA, is a document reporting selected analytical results for a defined sample. It can support a narrow quality question. It cannot, by itself, prove that every lot is identical, that a product is sterile or clinically safe, or that a material is suitable for human use.

Start with traceability

The first question is whether the document can be connected to the material under discussion. Look for a lot or batch identifier, sample identifier, date, supplier or submitter where appropriate, and a clear statement of what was tested. A generic certificate without a traceable lot has limited value. A result from a different batch, old inventory, or an unspecified sample should not be treated as proof for a current item.

Traceability also includes chain of custody. Who provided the sample? Did the laboratory receive a sealed, representative sample? Was the sample selected by the seller, a customer, or an independent party? A COA rarely answers every custody question, but the absence of information should reduce the certainty of the conclusion rather than invite assumption.

Identity is not purity

Identity testing asks whether the analyzed material is consistent with the expected analyte. Methods may include mass spectrometry or chromatographic comparison, depending on the question and laboratory. Purity asks a different question: how much of the measured signal or material corresponds to a target under the stated method. A high purity percentage does not establish that the identity assignment is correct in every respect, nor does identity establish the absence of every impurity.

Method details matter. A result should state the analytical technique, units, acceptance criterion if one is used, and result for the tested sample. Readers should avoid treating a single percentage as a universal score. The method may not detect every relevant impurity, degradation product, residual solvent, or contaminant. Understanding endotoxin claims

Endotoxin is a topic that demands precision. A reported endotoxin value should identify units, method, sample, and detection limits. It is not synonymous with sterility, and it is not a complete safety assessment. A result from one test cannot prove freedom from every microbial, particulate, chemical, or handling-related issue. It also does not authorize administration to people.

USP standards and laboratory practices provide frameworks for validated analytical work, but readers should not claim compliance merely because a document uses technical terms. Ask whether the report identifies the compendial or validated method, controls, date, and interpretation. If a claim is important, a qualified analytical professional can explain whether the method and scope fit the intended research question.

Red flags in presentation

  • No lot number or test date.
  • A cropped image that hides method details or laboratory identity.
  • A broad “99% pure” claim with no stated technique, chromatogram, or scope.
  • A certificate used to support medical outcomes or personal-use instructions.
  • Results that appear copied across unrelated products or batches.

These are reasons for further questions, not automatic proof of misconduct. The disciplined response is to request clearer documentation or reduce confidence, not to invent conclusions.

What a COA can responsibly support

At most, a credible COA may support a statement such as: a named laboratory reported a specified result for a defined sample using a stated method on a given date. That is meaningful in research quality review. A practical reading sequence

Read the document from identifiers to methods to results to limitations. Match the lot. Identify the exact analyte. Note the method and units. Check what was not tested. Then ask whether the claim being made is no broader than the report. Keep a copy of the document and the date if it is relevant to a research record. Do not use a COA as a substitute for validated institutional procedures, product approval, or clinical evaluation.

Comparing documents over time

Several certificates should not be averaged into a claim of consistency. First establish whether they concern comparable lots, methods, and specifications. A changed method, reporting threshold, sample preparation, or laboratory can make two percentages appear comparable when they are not. A repeated document template is not the same as repeated measurement. The useful record is a dated collection of source documents with their individual limits preserved.

Formal quality systems define acceptance criteria before testing and record deviations rather than selecting favorable results afterward. A public reader does not need to recreate a laboratory system, but can use the same habit: ask what was planned, what was measured, and what remains outside the result. That discipline is more informative than an isolated certificate image.

Questions for an analytical laboratory

If a result will influence a consequential research decision, ask a qualified laboratory about sample receipt, method suitability, calibration or reference standards, reportable range, uncertainty, and whether the reported analyte was distinguishable from likely related substances. The aim is not to demand a universal pass statement. It is to learn whether the method answers the exact question. Medical suitability is a different question entirely.

Bottom line

A careful COA reader values specificity over slogans. Identity, purity, and endotoxin are distinct analytical questions. Each result is limited by its method and tested sample. This page is for research literacy and does not provide purchasing, preparation, or personal-use guidance.

Build the traceability chain before reading the number

A useful review starts outside the result table. Match the report identifier to the lot, verify when the sample was received and tested, note who selected and submitted it, and record whether seals or receipt condition were described. A seller-selected vial can still generate a valid measurement for that vial, but it may not represent unsampled inventory. A report hosted as an editable image without a laboratory-verifiable identifier deserves less confidence than a complete, dated document whose issuing laboratory can confirm its scope.

Chain of custody does not prove quality; it makes the tested object identifiable. If custody is missing, readers cannot know whether the result belongs to the item being promoted. If custody is present, the conclusion still stops at the sample, methods, and attributes examined.

Orthogonal methods answer different parts of identity

Chromatography can separate components under stated conditions, while mass spectrometry can provide mass-related evidence about an analyte. Neither method should be described as a magical authenticity test. Retention time, detector choice, reference standards, ion patterns, sample preparation, and data interpretation affect what can be concluded. Agreement between appropriately chosen methods may strengthen an identity assessment because the methods rely on different measurement principles, but the report must show enough context to understand that agreement.

Purity percentages require equal restraint. Area percent from a chromatogram is not automatically mass fraction, content per vial, or absence of every contaminant. Co-eluting substances, detector response, integration choices, and method scope can affect the reported value. Quantity or assay, residual solvents, water, counterions, bioburden, sterility, and particulate matter are separate questions unless the report explicitly addresses them.

Read endotoxin as a method-specific result

An endotoxin statement should identify units and their denominator, method, dilution or interference controls where relevant, acceptance criterion, and reporting limit. “Pass” without those details is difficult to interpret. Endotoxin testing is not sterility testing: a sample may meet one stated endotoxin criterion while other microbial, particulate, or chemical risks remain unexamined. A low or non-detected result also cannot be generalized to other lots or to handling after the tested sample left the laboratory.

A compact red-flag checklist

  • The product name is visible but the lot, report number, or test date is missing.
  • The laboratory logo is shown while contact details, authorization, or complete pages are cropped away.
  • Identity, purity, quantity, sterility, and safety are compressed into one unexplained percentage.
  • The report provides a result but no method, units, specification, or detection/reporting limit.
  • The same chromatogram or exact values appear across supposedly different lots.
  • A vendor turns an analytical result into a claim about effectiveness, dosing, or suitability for people.

A red flag calls for clarification or reduced confidence; it is not permission to invent an accusation. The sound conclusion states exactly what cannot be verified.

Document review is not laboratory accreditation

Public readers can examine completeness and internal consistency, but they usually cannot audit instruments, analyst competence, validated procedures, data integrity controls, or the laboratory’s accreditation scope. Even an accredited laboratory is accredited for defined activities, not every test it might list. When a consequential research decision depends on a result, the issuing laboratory and qualified quality personnel are the appropriate sources for method suitability and uncertainty. The COA remains one record within a quality system, not a universal product verdict.
